Although one could conceivably drive from Boston to New York in as little as 3½ hours by just barreling down the Interstate, there is certainly a lot to do if you have the time and wander a bit. For the most part some (but only some) of the best sights are along the coast, so that's what I'd recommend as a general route. Highlights would include Plymouth Rock, Cape Cod, maybe a day trip out to Martha's Vineyard, Newport and the 'cottages', Mystic Seaport, and yes there is a real Mystic Pizza. Then depending on where you're going in New York City, you could either continue to follow the coast and walk Yale's campus, or take the ferry from New London over to Orient Point on Long Island and drive in that way.
