Driving from MSP to Orlando is not something you should even consider doing in one day. In fact, since you're looking at 27 hours on the road to cover the distance, its not even physically possible!
The most direct route would be I-94 to I-39 to I-74 to I-55 through Illinois, then I-24 through Nashville to I-75 on to Florida. That's about a 1550 mile trip.
3 days would really be a recommended pace. You could cut it down to 2 very full days, but then you'll want to keep your activities pretty low key on your first day in Orlando so you can rest up from the fairly grueling drive. Trying to drive to Orlando in 2 days and then trying to cram in a Theme Park the next day would be asking for a disaster.
