How much time do you have for this trip, and exactly what are you looking for. There is no such thing as a generic 'best' way to go.
Get out your map (not electronics) and check which roads/routes go that way. You can take the interstate the whole way, and pay the tolls. If you'd prefer a more sedate pace than interstates, PA59 will take you through the Allegheny NF to US219, which takes you all the way to Buffalo and Niagara. Or if you would like a more scenic route take I-86 to Maplehurst and NY16 to Buffalo.
These are just three I have picked off the map in front of me as I type (and have travelled in the past), but there are countless other routes to choose from, depending on what it is you are looking for in line with the interests of those who are travelling. Routes which you will not see on electronics. That is why good detailed maps are so valuable during planning a trip, and essential while you are on the road.
If you don't have some good detailed maps or a road atlas, go to AAA and get one there. If you are a member maps are free.

The trip your planning can be pretty straightforward - as it's only about 400 miles.
However, you really haven't told us enough for anyone to give you any useful suggestions. First of all, we don't even know what kinds of things you are looking for help with. Beyond that, How long do you have for this "quick" trip? What kinds of things are you interested in seeing or doing? Anything beyond the falls you know you do or don't want to do? Do you have your passports so you can cross to the Canadian side?
