There is no such thing as a one sized fits all "best route." Going the most direct route, you can make this trip in 2.5 days, but if you've got more time there are lots of detours you could take, like going through Yellowstone, or spending some time in Colorado. However, without knowing anything about you, your interests, the time you've got available, and the ideas you've already come up with, we simply can't be any more specific than that.

Are you riding, towing, or neither as you head to Sturgis? I've never been to the rally, but I've passed by coicident to the arrival weekend and the departure weekend. The highways (and gas stations, restaurants, motels, and campgrounds) get very crowded with ralliers for several hundred miles in every direction.
For scenery, I'd look at I-15/I-70/I-76 to northeasternmost Colorado, then north up US 385 through the Nebraska panhandle into SD. Sidney, NE (home of Cabela's), Alliance, NE (Carhenge), Chadron, NE (Museum of the Fur Trade/Sandoz High Plains Center), and Crawford, NE (Fort Robinson State Park) are all nice places for a visit. Even though they're off of the main beaten patch, I seriously doubt you'll be alone.
