Best can mean a million different things, and without any information, its impossible to know what might be best for you.
The fastest route would be I-65 to Nashville, I-24 to Chattanooga, and I-75/FL Turnpike to Orlando. That's about 1200 miles and is two very full days on the road. Of course, you could use any of the several online mapping programs and also play around to find something that might be more to your liking.

There is no "best" route unless you are simply asking how to get there the fastest and safest with no sightseeing. If this is the case:
I-57/I-24/I-75/Florida Turnpike
My mapping program says that the route Michael has suggested is close to an hour longer. I would tend to believe it - I-57 will avoid most of the I-80 congestion south of Chicago and both the Indianapolis and Louisville metros.
This will be a long 2 day drive, suggested halfway point for overnight is Chattanooga. With normal food, fuel, and bathroom stops you will be on the road about 12 hours a day.
Taking I-57 adds about 20 miles and it's certainly another reasonable option, but I think Streets and Trips is really over exaggerating the time difference.
Considering the shorter distance and the fact that Indiana has a higher speed limit than Illinois, the only way I see a significant time difference is if you hit an extremely heavy amount of traffic in Chicago and Indianapolis. For the shorter route here to take a full hour longer doesn't seem very logical.
