You could make the drive from Toronto to Philadelphia in a single day by sticking to the Interstates and not stopping for anything other than the most basic food, fuel and rest stops. By taking two days you will have more than enough time to take more relaxing 'back' roads, stop and see a few scenic sites up close, and spend an overnight on the way.
The basic idea is to just follow a different route from Buffalo to Philadelphia which, by its very nature, will not only take you through some very scenic areas of the Niagara Escarpment, the Allegheny Plateau, and the Appalachian Mountains, but will also save you a few miles of driving. You'd strike out on this other route at Batavia NY, taking NY-63 down past Letchworth State Park to Geneseo, and then use I-390/I-86 to Corning NY and US-15. US-15 follows the Susquehanna River down through central Pennsylvania past Williamsport to Harrisburg, near Hershey, and from there you can follow some two-lane roads such as PA-230 and PA-772 through Amish country and picturesque towns such as Lititz into the Philadelphia area.
Following such a route Mansfield PA, on US-6, would be about half way.
