From one corner of the country to the other, you can almost choose any route you want. It's going to be over 3000 miles anyway you do it.
So what are you interested in? Scenery? Nightlife? History? National Parks? All of the above? Maybe that can help narrow it down!
Like I said, I'll be doing it with my dog, so nightlife is probably out of the question. I'm thinking more scenery stuff. Unusual roadside stops. I did Florida to Oregon last year, but had my pregnant wife with me then, so needless to say we were'nt stopping for the sights. Unless you count bathroom breaks.

You can then look at the map and find the scenic national parks that are along your route.

Rte 1: Ast-Crater Lake-Yellowstone-Grand Teton-Little Bighorn-Deadwood/Black Hills-Wounded Knee-Amana Colonies (IA)-Springfield (IL)-Fort Donelson National Battlefield (TN)-Nashville-Tampa. 4100 miles, 69 hours driving (estimated). Add the Great Smokey Mtns for a little extra.
Rte 2: Ast-Crater Lake-Yosemite-Kings Canyon-Death Valley-Zion-Bryce-Capitol Reef-Durango-Lincoln (NM)-Carlsbad Caverns-Big Bend Natl Park (TX)-The Alamo-New Orleans-Tampa. 4700 miles, 87 driving hours, approx.
