Without going too far afield (so you can spend your time seeing things rather than driving to them) you can actually see quite a bit in western New York, northwestern Pennsylvania, an northeastern Ohio. Before even leaving Canada, check out Niagara-on-the-Lake. Then in western New York, there's Letchworth State Park, and the entire area is home to several great wineries. For a bit of 'fun' continue down to Jamestown NY and the Lucy Desi Museum, and for a bit of the spiritual and the arts check out the Chautauqua Institution. Or you can wander through the historic homes and museums of the Fort Leboeuf Historical Society in Waterford PA.

If you're interested at all in history, Lockport on the Erie Canal isn't all that far off the beaten track. Or, in Buffalo area, if you like architecture,Frank Lloyd-Wright's Darwin Martin House.
Another historical point, Pres. Teddy Roosevelt's inaugural place, is located near Buffalo. Or how about the Albright-Knox Art Gallery?

Another option to consider is re-routing entirely via Detroit. I don't know much about things to see along the 401 in Ontario, but the Henry Ford in Detroit is an amazing museum, and Perry's Victory Memorial at Put-In-Bay, OH is a unique National Park Site. Whether you visit the memorial or not (requires a boat ride from Catawba Island), I would take the toll-free OH-2 between Toledo and Sandusky, and (depending on time) ride US-6 along the lake back to Cleveland.
