Well, the term "best" is always a difficult one because it can mean so many different things. I will say when I make the trip, I'll typically cross into Canada in Michigan (Either in Detroit or north at Sarnia depending upon my mood) and go across that way. Any of those routes will save you about 30 miles, but more importantly, it will save you a significant amount in tolls.
In any case, you're looking at a trip that's about 550 miles and will take you about 10 hours before you start making more than the bare minimum of stops.
