The shortest route is basically I-79 from Erie South to I-77 (US-19 is a shortcut through West Virginia) to I-95 in South Carolina.
You are looking at 2 very full days of at least 12 hours each. Hillsville, VA is slightly over halfway, and at 650 miles, its really maximum you should be looking at doing on a family trip anyway.
