I'm planning to driving from Death Valley (Furnace Creek) to Yosemite Park (Stoping at Yosemite Falls Lodge) this Sep 09. Not sure what the best route is and how long this will take.

There isn't only one route, but the most direct, obvious, and scenic choice is to take Tioga Pass through Yosemite across the Sierras. I'd say you're looking at 5-6 hours, depending upon how often you stop to look at the many possible sites.
Yep, it can't be beaten, take the 190/136 to Lone pine and head North to Lee vining on 395 and then the Tioga pass [120] into Yosemite. With 300 miles and a lot of sight seeing opportunity's I would get an early start and expect a long, but very enjoyable day.
